培养条件对Coriolus Versicolor菌分泌漆酶的影响

摘要

采用云芝菌(coriolus versicolor)摇瓶发酵生产漆酶,对主要的影响因素进行了优化。讨论了碳源、氮源、铜离子浓度、诱导剂、表面活性剂等各种因素对杂色云芝菌分泌漆酶能力的影响,获得了优化的培养基组成:土豆汁20%,葡萄糖20 g/L,蛋白胨4 g/L,CuSO40.5 mmol/L,KH2PO43 g/L,MgSO41.5 g/L,ZnSO450 mg/L,MnSO450 mg/L。结果表明,表面活性剂Tween-80的加入能显著提高漆酶的分泌,最佳诱导剂为ABTS。在最优条件下发酵13 d,漆酶活力可高达92.2 U/mL。

Abstract

Effects of Producing Laccase on Coriolus Versicolor by Different Fermentation ConditionsFANG Hua,HUANG Jun,CHEN Zhao,ZHANG De-qing,DING Li-yun (Key Laboratory of Fiber Optic Sensing Technology and Information Processing of Ministry of Education,Wuhan University of Technology,Wuhan 430070,China)The main factors affecting laccase production by coriolus versicolor in liquid shake-flask fermentation were researched.The effects of pH,temperature,carbon sources,nitrogen sources and copper concentration on producing laccase of coriolus versicolor were researched.The optimal fermentation condition for laccase production were glucose 20 g/L,peptone 4 g/L,trace metal CuSO4 0.5 mmol/L,KH2PO4 3 g/L,MgSO4 1.5 g/L,ZnSO4 50 mg/L,MnSO4 50 mg/L,Tween-80 4 g/L,pH 5.0,and incubation with 130 r/min at 28 ℃ for 13 days.The capabilities of producing laccase were evidently enhanced with the addition of Tween-80 and ABTS.Laccase activity was 92.2 U/mL in the optimal fermentation condition.coriolus versicolor;laccase;fermentation condition;relative activity
